Knoppix 5.3.1 di-upadate mengemas Kernel Linux 2.6.24.4 dan KDE 3.5.9 berikut compiz-fusion sebagai 3D-Windowmanager. Opsi untuk menjalankan generasi terbaru KDE-4 juga disediakan sebagai Bootoption, pilihan saat awal menjalankan LiveDVD.
Sebagaimana telah disebutkan sebelumnya, mulai versi 5.3, Klaus menyertakan dukungan untuk Tunanetra dengan sistem desktop ADRIANE (Audio Desktop Reference Implementation and Networking Environment) yang dikembangkannya berikut Screenreader Orca.
Program untuk instalasi atau menanamkan Knoppix di harddisk '0wn' atau 'Zero Work Needed' pada edisi ini mendapat peningkatan signifikan. Driver yang mendukung penulisan di partisi windows ntfs-3g juga ditingkatkan.

ADRIANE, Audio Desktop with text-to-speech and braille support, first release (not fully complete yet), bootoption "adriane", see http://knopper.net/knoppix-adriane/index-en.html. Known bug: even theenglish version talks in german, this can be fixed by removing all the language-related lines in /etc/adriane/adriane.conf, and restarting the screenreader with "adriane-screenreader" from the shell.
